- W1567245911 abstract "In the 1960s, Godfraind and his colleagues [1 2 3 4] studied the mechanism of action of polyvalent antagonists, i.e. agents that block in a similar concentration range the vasoconstriction evoked by several agonists acting on different receptors. They concluded that those agents, including various diphenylpiperazines, interfere with a common calcium entry process activated by the various vasoconstrictor agonists, and showed that inhibition of calcium entry in stimulated cells is a mechanism of drug action. An important observation was that these agents are able to inhibit the contraction evoked by calcium in depolarized smooth muscle in a manner similar to the action of antagonists observed in receptor studies; this supported the use of the term “calcium antagonist” coincidentally used by Fleckenstein to describe the effects of verapamil and prenylamine on cardiac tissue[5]." @default.
