- W1567682067 abstract "Abstract A method for the degradation of 14C-labeled glycerol in which all carbons are converted to CO2 is deseribed, using commercially available enzymes. Glycerol is enzymically phosphorylated and oxidized to phosphoglyceric acid in a one-step procedure. The conversion is nearly quantitative. Phosphoglyceric acid is hydrolyzed to glyceric acid. Glyceric acid is oxidized by periodate, yielding CO2 from C-1, formaldehyde from C-3, and formic acid from C-2. Formaldehyde and formic acid are separated and oxidized to CO2. The CO2 is converted to BaCO3 for the determination of specific activity. The procedure employs simple equipment, is rapid, and lends itself to simultancous processing of several samples. A procedure for glyceric acid assay based on oxidation to glyocylic acid is deseribed. Glyoxylic acid is assayed with lactic acid dehydrogenase. We have observed that phosphate is required for the complete oxidation of glycerate by periodate. In the absence of phosphate, in a pH range from 2 to 8, glyceric acid is oxidized rapidly to glyoxylic acid, which is oxidized further at a slow rate." @default.
