- W1569241502 abstract "This chapter describes a series of methods developed from studies on cytochrome P450 terpenoid hydroxylases involved in resin acid and taxol biosynthesis in gymnosperms, including pine, fir, and yew species. Woody tissues carry out many of the same cytochrome P450-dependent primary metabolic reactions as their herbaceous counterparts and are often rich in secondary metabolites, the biosynthesis of which involves cytochrome P450-catalyzed steps. High quality microsomes may be extracted from woody tissues using a variety of buffers. However, all these media should contain agents that protect cytochrome P450 from inactivation and promote efficient recovery of this enzyme. The techniques range from protocols for small-scale survey work, in which tissue can be completely pulverized, to intermediate-scale preparations involving shearing of tissue and hammer milling, to large-scale isolation requiring a substantial chopping mill. All these operations are performed at subambient temperatures, employ buffers designed to minimize the influence of deleterious coextractives, maintain membrane integrity, and are adaptable to a wide spectrum of woody tissues." @default.
