- W1569288872 abstract "This chapter analyzes the reactive oxygen species in cell death. The ability of oxidants to trigger apoptosis depends on the extent of the oxidative stress. Higher oxidant concentrations lead to necrosis and the “window” for induction of apoptosis may be small, depending on the cell type and the oxidant investigated. Mammalian cells possess multiple sources of reactive oxygen species (ROS), including, among others, xanthine oxidase, mitochondrial NADH oxidoreductase, microsomal NADPH oxidoreductase, and plasma membrane-bound NADH/NADPH oxidases. Evidence suggests that plasma membrane-associated oxidases may provide one source of ROS associated with cell resistance to triggers of apoptosis. NADH and NADPH oxidases may provide candidate sources of superoxide anion production associated with cell resistance to apoptosis. This chapter discusses the role of NADH and NADPH oxidases in the regulation of sensitivity to apoptosis. The chapter provides a method to measure the redox state of the active site cysteine of caspases. This chapter discusses the method based on the ability of iodoacetic acid (IAA) to react with reduced but not oxidized thiols and the visualization of IAA incorporation by its ability to shift the mobility of the target protein on an isoelectric focusing (IEF) gel." @default.
