- W1569392810 abstract "Microwave discharge activation of tritium gas in a cycling system has resulted in a tritium labeling method which is particularly well suited to the preparation of high specific activity, biologically active tritiated peptides. Most of the tritiated impurities are apparently associated with fragmentation of the peptide, the polymeric nature of which permits their facile separation. In most cases, the remaining impurities differ from the parent compound sufficiently to allow purification by ion-exchange chromatography. Amino acid analysis of purified peptide reveals that at least 90% of the peptide tritium can be accounted for by the tritium in the component amino acids and the tritium exchanged during base and/or acid hydrolysis. Specific activities from 1 to 25 Ci/mmole can be obtained with preservation of full biologic activity in the case of [3H]ACTH. Similar specific activities have also been obtained in experiments with ovine luteinizing hormone in which biologic activity was maintained. Precise new bioassay methods and improved analytical techniques will make it possible to determine whether tritiated protein molecules retain biologic activity and function." @default.
