Deney sonunda, tum deneklerin kalp ornekleri al›narak ›fl›k ve elektron mik- roskobu ile incelendi; dokudaki katalaz, glutatyon, glutatyon-S-tranferaz, glutatyon peroksidaz ve protein oksidasyon aktivitelerine bak›ld›. Bulgular: Kontrol ve nikotinamid grubu deneklerde kalp dokusu normal yap›dayd›. Ucuncu grupta, iltihap hucre infiltrasyonu, miyokard lif- lerinde duzensizlik vard›. Dokuda katalaz (CAT), glutatyon (GSH), glutatyon-S-tranferaz (GST), glutatyon peroksidaz (GSH-Px) aktivitesi azal- m›fl, protein oksidasyonu artm›flt›. Doksorubisinle birlikte nikotinamid verilen grupta GSH-Px, CAT, GSH, GST’nin yukseldi¤i, histopatolo- jik olarak nikotinamidin kardiyomiyopatiyi onledi¤i gozlemlendi. Sonuc: Antioksidan olarak kullan›lan nikotinamidin doksorubisin kardiyotoksisitesinin azalmas›na yard›mc› oldu¤u ve ileride yap›lacak ge- nifl kapsaml› cal›flmalarla klinik uygulamalara katk›da bulunabilece¤in duflunuldu. Anahtar kelimeler: Antineoplastik ajan, doksorubisin, nikotinamid, deneysel kardiyotoksisite, antioksidan Cerrahpafla T›p Derg 2008; 39: 7-14 Effect of nicotinamide on doxorubicin induced experimental cardiotoxicity Abstract Objectives: We investigated the morphologic changes showing cardiotoxicity caused by use of the doxorubicin which is an antineoplastic agent, and the protective effect of nicotinamide. Methods: We assigned thirty Wistar Albino male rats were randomly to four groups: Group 1 (Control) had %0.9 NaCl for 7 days, group 2 had Nicotinamide 200 mg/kg/day for 7 days, group 3 had single dose injection of Doxorubicin (20 mg/kg), group 4 had combination of Do- xorubicin and Nicotinamide intraperitoneally. At the end of the experiment, the hearts were removed for light and electron microscopic in- vestigations. We determined the tissue activites of catalase, glutathione-S-transferase, glutathione, glutathione peroxidase. Results: Control and Nicotinamide rats showed a normal heart tissue morphology. The rats in group 3 exhibited disorganization of the myo- cardial fibres, and infiltration of inflammatory cells. The rats in group 4 displayed a significant decrease in morphological damage, with the disappearance of inflammatory cell infiltration. Conclusion: Our findings suggested that the use of antioxidants of Nicotinamide may help to reduce Doxorubicin cardiotoxicity, thus con- tributing to clinical applications.
