- W1570325020 abstract "613 Objectives The ROLL technique, developed at our Institute, consists in the inoculation of particles of colloidal human albumin (HA) aspecifically labelled with 99mTc directly into the non-palpable breast lesion to achieve a precise localization before surgery. Methods The short half-life of 99mTc does not allow to plan surgical procedures on several days and to carry out the localization as an outpatient exam thus limiting patient accessibility to this methodology. We have developed a microdispersed system (HAC) able to complex radionuclides such as 111In and 177Lu. The new system is produced from commercial HA and is obtainable in a defined particle size. HA was initially purified and mixed with p-SCN-Bn-DOTA. The mixture was heated and the solid phase thus formed was purified by ultrafiltration and then lyophilized. Evidence for the formation of HAC as a new chemical entity was provided by MALDI-TOF mass spectrometry. Results The degree of substitution (DS) was influenced by reactant molar ratio, pH, temperature and reaction time. In the optimized conditions, DS averaged 5 nDOTA/nHA, coherent with the estimation obtained by MALDI-TOF. Radiolabelling tests using 111In and 177Lu showed that HAC can withstand to the high temperature required for a high labeling yield. Stability studies of 177Lu-DOTA-HAC up to 6 days at 37 °C either in PBS or human plasma showed no evidence of radiolysis or biodegradation of the conjugate. Conclusions In conclusion, we have developed a new HA conjugate suitable for labeling with 111In and 177Lu which can be produced as a sterile and lyophilized product with the aim to expand the application of the ROLL procedure." @default.
