OBJECTIVE: To prepare a new poly-asparagine derivates, namely succinyl-α, β-poly(2-hydroethyl)-D,L-aspartamide (PHEA-Suc) and galactosyl-PHEA-Suc (Gal-PHEA-Suc), and to explore its characterization and cytotoxicity in vitro. DESIGN, TIME AND SETTING: The in vitro trial was performed in the State Key Laboratory of Pharmaceutical Biotechnology, School of Life Science, Nanjing University (Nanjing, Jiangsu, China) from June 2005 to June 2007. MATERIALS: L-aspartic acid, anhydrous N, N-dimethylformamide. lactobionic acid, fluorescein isothtocyanate, dimethyl sulfoxide and MTT were purchased from Sigma Company (USA); Human HeLa cells and HepG2 cells were offered from Shanghat Cell Bank; New-born calf serum was a product of Hangzhou Sijiqing Biocompany; Trypase was purchased from Hyclone Company (USA). Other reagents were domestic analytical pure. METHODS: Infrared spectrum, nuclear magnetic resonance, elemental analysis and molecular mass determinatson were all conducted to investigate the characterization of the material. Tetrazolium bromide colorimetric assay was employed to detect the cytotoxicity of PHEA-sec and Gal-PHEA-suc. MAIN OUTCOME MEASURES: The structure and cytotoxicity of poly-asparagine derivates carrier. RESULTS: The structures f PHEA-suc and Gal-PHEA-suc were identified using infrared spectrum and nuclear magnetic resonance. Elemental analysis displayed that the succinylation degree of PHEA-suc and Gal-PHEA-suc were 43% (mol/mol), galactonylation degree of Gal-PHEA-suc was 7.6% (mol/mol). Results of in vitro cytotoxicity showed that, both PHEA-suc and Gal-PHEA-suc exhibited few effects on inhibiting the growth of BeLa cells and HepG2 cells. There was no effect action at a concentration of less than 500 mg/L. CONCLUSION: Poly-asparagine derivates carriers PHEA-suc and Gal-PHEA-suc have low cytotoxicity with-suc active group and-gal target group, so they can be used as ideal target drug carriers." @default.
