- W1573485805 abstract "Publisher Summary This chapter describes the applications of yeast in the discovery of human gene function. The convenience of handling and the ease of both classical and recombinant genetic manipulation have made yeast Saccharornyces cerevisae an invaluable model system in which both metabolic pathways and fundamental cellular processes, such as the cell cycle, DNA repair, transcription, translation, and protein secretion, can be dissected. Subsequent analysis of these pathways in higher eukaryotes has demonstrated that, despite a large evolutionary distance, yeast and humans exhibit a remarkable conservation of gene function. A consequence of this conservation is the ability to identify the functional homolog (ortholog) of a yeast gene from another eukaryote. The chapter describes approaches used to identify human homologs of a yeast gene by database homology searching and yeast genetic screens suitable for the functional isolation of human genes that lack a counterpart in yeast. The analysis of the human gene function by comparative yeast genomics is presented in the chapter. It also discusses the identification of human genes by genetic selection in yeast." @default.
