- W157495220 abstract "This chapter discusses rate-limiting factors in HeLa cells in relation to metabolism of intact cells. In an experiment described in the chapter, the profile of glycolytic enzymes in HeLa cells was, on the whole, remarkably similar to that of ascites tumor cells. Hexokinase and phosphofructokinase were relatively low, whereas phosphoglycerate kinase was about 100 times as active. The over-all glucose metabolism was also similar. The rate of glucose utilization in fortified homogenates was much faster than in intact cells, indicating that the enzymes were present in excess. High phosphate concentration in the medium resulted in a marked stimulation of glucose utilization by the intact cells. In contrast to ascites cells, the HeLa cells exhibited endogenous fermentation as a result of the breakdown of glycogen. While lactate production from glucose showed no Pasteur effect, the production of lactate from glycogen was much faster anaerobically than aerobically. Addition of both P and AMP to the cells markedly stimulated the endogenous formation of lactate. Measurements of phosphorylase activity in cell-free extracts of HeLa cells revealed an activity considerably lower than that of other glycolytic enzymes." @default.
