This chapter focuses on the application and advantages of flow cytometry to measure virus neutralization. Virus neutralization assays are designed to measure a reduction in virus infectious titer mediated by exposure to antibody. The chapter presents basic methods for an HIV-1 flow cytometric neutralization assay. Antibodies bound to viral determinants can interrupt viral infection of a cell by several mechanisms, including the inhibition of cellular attachment, virus-cell fusion, and uncoating after viral entry. The ability to make accurate and precise measurements of antibody-mediated virus neutralization is important for the evaluation of mechanisms of neutralization and for the assessment of antibody responses elicited by immunization." @default.
