- W158011 abstract "The objective of this study was to determine if DNA damage increases as a function of age in post-mitotic cells. A (/sup 32/P)-post-labeling technique was used to measure the relative level of alkylated bases in mouse myocardial DNA. DNA was purified from heart tissue of male, C57BL/6nia mice at different ages. Samples were digested to deoxynucleoside-3'-monophosphates with micrococcal nuclease and spleen phosphodiesterase and labeled with (..gamma..-/sup 32/P) ATP and T4 polynucleotide kinase. The resultant nucleotide diphosphates were converted to deoxynucleoside-5'-monophosphates with nuclease P1. Normal nucleotides were then separated from alkylated nucleotides by two-dimensional thin-layer chromatography. Adducts were detected by autoradiography and quantified by liquid scintillation counting. A modified nucleotide was found to increase more than 50-fold between 2 months and 39 months. The greatest increase occurred in senescent mice, older than 17 months. This modified nucleotide comigrated with 7-methylguanosine-5'-monophosphate in the two-dimensional chromatography system. Furthermore, this nucleotide was induced by N-methyl-N-nitrosourea alkylation of poly(dG-dC) x poly(dG-dC) but not poly(dA-dT) x poly(dA-dT). The results are consistent with an age-related increase in the steady-state level of DNA in mouse heart DNA." @default.
