- W1587648980 abstract "In this report we examined the production of γ-aminobutyric acid (GABA) from glutamate using Aspergillus oryzae cultured in a liquid medium as enzyme (glutamate decarboxylase; GAD) sources, and separation of the reaction mixture into GABA and glutamic acid using electric dialysis. Furthermore, we examined the optimum conditions for obtaining A. oryzae with the highest GAD activity in order to increase the GABA production rate.We found that the liquid medium for growing A. oryzae with the highest GAD activity was best at pH 5. 0 and included a PLP of 50 μM.When we put 500 mM of glutamate and O. 5 mM of PLP into the stirring batch reactor controlled under pH 5. 3-5. 5, which included pellet form mycelium of A. oryzae (dry weight 6. 0 g) cultured in the liquid medium and treated under a low temperature (4°C) to increase the GAD activity, the concentration of GABA produced was 340 mM after 100 minutes of reaction time. From these findings, the conversion rate from the substrate to GABA at a reaction time of 200 minutes seemed high and was calculated as 80%. The production rate was extremely high with 8. 5 mmol GABA/g Aspergillus mycelium/hr (0. 9 g/g/hr). On the basis of GAD stability, the optimum temperature in the reactor was apparently 37°C. Electric dialysis using an ion-exchange membrane proved to be effective in separation of the reaction mixture into GABA and glutamic acid." @default.
