When measuring ROS in cells and mitochondria, the chapter emphasizes the need for thorough understanding of results obtained and their interpretation and describes the use of luminol as a detector of radicals formed during the reaction of xanthine oxidase (XO) with hypoxanthine (HX). It was reported that the light produced by luminol and XO/HX was directly related to the velocity of the XO/HX reaction, and this system could be used as a general test for oxidizing radicals. The chapter presents several processes for measurement of ROS in cells such as electron spin resonance spectroscopy, fluorescence dye techniques for the determination of cellular ROS, the determination of ROS using hydroethidine and fluorescence activated cell sorting analyses, and spectrophotometric methods for determination of ROS, as well as the process of measurement of mitochondrial ROS. In the presence of horseradish peroxidase (HRP), amplex red reagent (10-acetyl-3, 7-dihydroxyphenoxazine) reacts with H2O2 with a 1:1 stoichiometry to produce a highly fluorescent resorufin product. Amplex red/HRP has been observed to detect levels of H2O2 produced by the NADPH oxidase and other oxidases." @default.
