Soo, Ophrys apifera Hudson, Ophrys ferrum- equinum Desf, Orchis coriophora L., Orchis laxiflora Lam., Orchis mascula (L.) L. ssp. pinetorum (Boiss et Kotschy) Camus, Orchis morio L., Orchis pallens L., Orchis purpurea Hudson, Orchis simia Lam., Orchis tridentata Scop., Platanthera chlorantha (Custer) Reichb., Serapias vomeracea (Burm.fil.) Briq. ssp. orientalis Greuter.) kullanilmistir. Farkli besin ortami bilesimleri (1/2 MS, MS, VW & DB, KC, KC-N), besin ortamlarina GA3 katkisi (0, 0.1, 0.5mg/l) yapilmasi ve inkubasyon sirasinda farkli aydinlatma rejimi uygulamalari (ilk bir ay veya ilk 3 ay karanlikta tutma ve ardindan fotoperiyodik duzene aktarma) yapilmis; bu uygulamalarin cimlenme, protokorm olusumu ve bitki gelisme oranlari uzerine etkileri incelenmistir. In vitro kosullarda cimlendirme ve gelistirme amaciyla yedi farkli deneme, dis kosullara aktarma amaciyla dort degisik deneme kurulmustur. Denemeye alinan tum turlerde cimlenme ve protokorm elde edilmis; ancak bunlardan ucunde cok dusuk oranlarda olmakla birlikte sadece sekiz tanesinde bitkiye donusum saglanabilmistir. Doku kulturundeki basari, basta genotip olmak uzere besin ortami, inkubasyon kosullari ve bitkinin yetisme kosullarindan etkilenmektedir. Bu nedenle interaksiyonlar on plana cikmaktadir. En yuksek bitki olusum orani Orchis morio’da % 89.88; Serapias vomeraceae’da % 57.65; Orchis coriophora’da % 48.35 ve Orchis laxiflora’da % 14.57 degerleriyle KC ortamindan; Dactylorhiza nieschalkiorum’da % 19.87’lik bir degerle 0.5mg/l GA3 ilave edilen KC besin ortamindan elde edilmistir. Dis kosullara alistirmada tam bir basari saglanamamis, bitkiler en fazla birkac ay icerisinde canliliklarini yitirmislerdir. Yumrularin bolunmesi ve ana yumrulu bitkinin dogal ortamina dikilmesi ile kurulan in situ denemelerde ise cikis meydana gelmemistir.Abstract Some of the important middle generation orchid species used for Salep production grows in Turkey. Western Black Sea region is also rich in terms of Salep orchids. However, they are under threat for extinction because of illegal hand picking from the nature. The aim of this study is to develop reproduction possibilities under in vitro and in vivo conditions. In order to germinate the seeds, not having the endosperm, under tissue culture asymbiotically 16 species were used. Those were (Anacamptis pyramidalis (L.) L.C. Rich, Dactylorhiza incarnata (L.) Soo, Dactylorhiza nieschalkiorum Bauman et Kunkele, Dactylorhiza romana (seb.) Soo, Ophrys apifera Hudson, Ophrys ferrum- equinum Desf, Orchis coriophora L., Orchis laxiflora Lam., Orchis mascula (L.) L. ssp. pinetorum (Boiss et Kotschy) Camus, Orchis morio L., Orchis pallens L., Orchis purpurea Hudson, Orchis simia Lam., Orchis tridentata Scop., Platanthera chlorantha (Custer) Reichb. and Serapias vomeracea (Burm.fil.) Briq. ssp. orientalis Greuter.) Different nutrition media compositions (1/2 MS, MS, VW & DB, KC, KC-N), GA3 addition to nutrition media (0, 0.1, 0.5mg/l), different lightning regime during incubation process (first one month or first three months under dark and transferred to photoperiodic arrangement) were carried out. Also, the effects of those applications to germination, protokorm formation and plant development rate were investigated. In order to germinate and develop the seeds under in vitro conditions, seven different trial areas and to transfer them to outdoors, 4 different trial areas were established. Germination and protokorm formation were obtained for all of the species taken under investigation. However, only eight of them turned into a plant and three of those were at very low rates. The success rate of tissue culture was affected especially by genotype, nutrition media, incubation conditions and plant growth conditions. Thus, interactions came to the fore. The highest plant formation ratio in KC nutrition media was found at Orchis moro as 89.88%, followed by Serapias vomeraceae as 57.65, Orchis coriphora as 48.35%, Orchis laxiflora as 14.57%. The highest plant formation ration in 0.5mg/l GA3 added KC media was found at Dactylorhiza nieschalkiorum as19.87%. Complete success could not be achieved for adaptation to exterior conditions; plants lost their vitality within couple of months. An output could not be obtained from in situ trials made by dividing the tubers and planting the main tuberous plant to its natural environment." @default.
