- W1593026472 abstract "Publisher Summary This chapter discusses the caveolin interaction with endothelial nitric oxide synthase (eNOS). Nitric oxide (NO) is an important signaling molecule involved in many biological processes and is synthesized by a family of calmodulin-activated NO synthase (NOS). The endothelial NOS (eNOS) is the NOS isoform responsible for cardiovascular homeostasis, including regulation of blood pressure, permeability, and angiogenesis but also anti-thrombotic activity, vessel remodeling, and cardiac contractility. Indeed, eNOS is not only expressed in the vascular endothelium but also in platelets, smooth muscle cells, and cardiac myocytes. Of the NOS isoforms, eNOS is unique in that it is dually acylated by the fatty acids myristate and palmitate, thereby leading to the membrane location of this enzyme. Since eNOS activity is acutely regulated by multiple extracellular stimuli and the NO produced is a very labile messenger molecule with primarily paracrine function, the site of NO synthesis has a major influence on its biological activity. In addition to the requirement that NO be produced in a site adapted to its gaseous nature and function, the cytotoxicity of high levels of NO implies a tight regulation of its production." @default.
