- W1594802364 abstract "Publisher Summary This chapter describes the procedures used to generate useful antibody reagents. It discusses how they are characterized and presents a critical evaluation of the methods and of the usefulness of the reagents that are obtained. Immunizing animals with synthetic oligopeptide antigens is less useful than using large fragments of CFTR expressed as fusion proteins. This is especially true in the production of hybridomas, where the combination of immunization with oligopeptides and an ELISA-based screening assay leads to the production of reagents that not only are not specific for CFTR, but give misleading cross-reactivities. On the other hand, the combination of immunization with fusion protein antigens and a dot-blot screening assay are highly successful. Employing crude bacterial lysates from a fusion protein expression system for both immunization and screening may be a rapid and cost-effective method for developing useful MAb reagents." @default.
