- W1601860624 abstract "The behavior of 10-day, embryonic, vertebral chondrocytes was examined in three different in vitro situations. Chondrocytes cultured in intact cartilages synthesized a sulfated polysaccharide which was indistinguishable from chondroitin sulfate. No net DNA synthesis was detected in such cartilages. When liberated from their matrix and cultured as pellets, chondrocytes continued to synthesize chondroitin sulfate and remained an essentially postmitotic population. Identical cells cultured as monodisperse populations on plasma clots, ceased synthesizing chondroitin sulfate and began to synthesize DNA and other molecules necessary for cell division. Concomitantly with the initiation of DNA synthesis, an unsulfated polysaccharide appeared in the system. This polysaccharide was not synthesized by similar cells in intact cartilages or in pellets. When the initially monodisperse cultures became “crowded,” net DNA synthesis ceased and the cells began making sulfated polysaccharides. Unsulfated polysaccharide continued to be synthesized in “crowded” cultures. Cells from liver and muscle were cultured under the same conditions as were chondrocytes. These populations synthesized both unsulfated and sulfated polysaccharides in dispersed and confluent cultures. It is concluded that (1) chondroitin sulfate synthesis and DNA synthesis appear to be mutually exclusive processes, (2) intimate cell association is optimal for the maintenance of the cartilage phenotype, (3) lack of association will induce replication together with loss of the ability to synthesize chondroitin sulfate, and (4) many cell types may react to an in vitro experience by dividing and synthesizing both unsulfated and sulfated polysaccharides." @default.
