- W1602807170 abstract "The need for the measurement of apolipoprotein A-I (apoA-I) levels in the plasma stems, from the fact that levels of high-density lipoprotein (HDL) and its major protein component, apoA-I, are inversely correlated with coronary artery disease (CAD). Several methods are available for the quantitation of apoA-I. Most methods involve either electrophoretic or immunochemical techniques. Gel-filtration and ion-exchange chromatographic methods have been used in this chapter for the isolation of apoA-I. However, for quantitation, these methods are very slow. A reliable, sensitive, and relatively rapid method for quantitation of apoA-I has been developed, using reversed-phase high performance liquid chromatography (HPLC). This involves the selective adsorption of apolipoproteins present in HDL onto a reversed-phase HPLC matrix and sequential release, using a suitable organic solvent gradient. The HPLC method is attractive for two main reasons: (1) there have been tremendous improvements in reversed-phase supports, and (2) because of the improvements in computer applications to chromatography-based estimation of proteins and other organic compounds, quantitation of peaks in a given HPLC profile has been very precise. High-performance liquid chromatography has been used for amino acid analysis, estimation of certain organic compounds in biological fluids and tissues, and protein sequencing. Such studies have been possible mainly because of high reproducibility, an essential criterion in any quantitation procedure. Despite this, HPLC has not been extensively used for the quantitation of apolipoproteins." @default.
