- W1604491005 abstract "Publisher Summary This chapter focuses on the vanilloid receptor 1 (VR1), which is considered as an integrator of noxius and inflammatory stimuli. The relationships between the various modalities sensed by polymodal nociceptors and the underlying transduction mechanisms were unknown until the cloning of a capsaicin sensitive receptor (VR1). It is now clear that the expression of VR1 confers on the cells the property of heat and capsaicin sensitivity, as well as the ability to respond to some mediators produced during inflammation. Emerging evidence also indicates that other membrane receptors can be involved in the transduction of noxious thermal stimuli. VR1 has a pivotal role in nociception because it integrates the response to thermal stimuli and a range of inflammatory mediators. The human homologue of VR1 has been cloned and characterized. This shows 85–86% identity and 92% similarity to the rat VR1 at the amino acid level, with the ankyrin repeat and transmembrane domains conserved. The N-terminus region shows the lowest homology with increasing divergence toward the N-terminus, while the C-terminus shows a greater degree of conservation. Differences in the amino acid sequence have been shown to result in divergent pharmacological properties of the rat and human VR1." @default.
