- W1604722597 abstract "This chapter discusses the use of retroviruses as insertional mutagens and the use of the non-acute transforming retrovirus MoMuLV for the induction of lymphoid malignancies in mice that has been shown to be a powerful method to identify new oncogenes and to gain more insight into the molecular wiring that underlies oncogene cooperation. Cancer is a multistep process and the ability to interfere with this process effectively and on all levels of progression depends on the knowledge of the mechanisms that make a cell a cancer cell. Some retroviruses bear oncogenes and allow a direct analysis of their activation. Other retroviruses can cause cancer without bearing the mutated genes that initiate or promote malignancy; they target the appropriate gene at the genomic level. Based on this ability, retroviruses have been used as tools to identify a large number of genes involved in various stages of tumorigenesis and to decipher the wiring of oncogene cooperation and the synergistic actions of oncogenes and suppressor genes. This will allow in future refining the understanding of genetic alterations required for malignant diseases. The degree of complementation and redundancy of different signaling pathways and any synergistic effects of oncogene cooperation in tumorigenesis will hopefully become evident." @default.
