- W1605884071 abstract "Bostryx scalariformis (Mollusca, Gastropoda) is a land snail species from central Peru. It is endemic to coastal ecosystem and inhabits mainly in sand formations. Alive individuals have only been found in Ancon, Pasamayo, Lachay and Cerro de Agua lomas from Lima department. This species has two morphotypes, which in turn are geographically isolated, one morphotype inhabits in Ancon-Pasamayo lomas and the other one in Lachay-Cerro de Agua. Habitat fragmentation and decreasing of population size could have lead to reduction of genetic diversity and to genetic differentiation between its populations. The aim of this research was to evaluate genetic diversity in B. scalariformis comparing it with both its geographical distribution and morphotypes, in order to know about its population genetic structure. A morphometrical analysis was performed with 200 shells from the four localities using 10 variables from the shell. Alive snails were used to extract total DNA from foot muscle and 16S rRNA gene was amplified by PCR. PCR products were sequenced and these data were used in phylogenetic and population analyzes. Morphometric analyses reveals differences – mainly in ornamentationbetween populations from Ancon-Pasamayo and LachayCerro de Agua. Multiple alignment of 61 sequences shows 224 conserved sites, 111 variable sites and 55 informative sites. Intraspecific phylogeny reveals two monophyletic clades inside B. scalariformis: Ancon-Pasamayo and Lachay-Cerro de Agua. Population analysis shows two groups of haplotypes corresponding to morphotypes. Network analysis shows also genetic divergence between morphotypes. Genetic differentiation resulted obvious between isolated morphotypes being the differences within Ancon-Pasamayo higher than Lachay-Cerro de Agua. Coastal desert has played a key role isolating populations and allows genetic differentation between morphotypes. Historical climate changes as ENSO had also influenced in maintaining genetic diversity in land snails. This work reveals B. scalariformis population genetic structure and the importance of lomas formations as hotspots of diversity. Conservation of these habitats is important because its key role in maintaining evolutionary potential of species an the high biodiversity present." @default.
