- W1607720944 abstract "Myxoviruses, including influenza A strains and Newcastle disease virus, are highly sensitive to inactivation by phospholipase C when grown in cultures of chick embryo fibroblasts but are resistant to this enzyme if propagated in the allantoic sac. This difference in enzyme susceptibility resulted from a true host-induced modification of the viral envelope. The addition of sphingomyelin to culture medium substantially increased resistance to phospholipase C of virus grown in vitro, suggesting that added phospholipid is incorporated into the host cell membrane which becomes the envelope of mature virions. Electron microscopic studies were carried out with synthetic phosphatide-sterol mixtures corresponding to the major lipid components of influenza virus. Under appropriate conditions these mixtures showed organized aggregates that resembled “spiked” virions of certain arboviruses and myxoviruses. It was further demonstrated that cholesterol interacts with unidentified, heat-labile material in crude phospholipase C preparations forming ringlike configurations. The data support the conclusion that phospholipase C can act on certain lipid-containing viruses to release host-derived cholesterol which forms these ring structures in the presence of the unidentified constituent." @default.
