Mullis in 1983, revolutionized the ability to perform biologic research and has become a technologic foundation in the molecular diagnostic laboratory. With its high sensitivity for detecting and copying a target region from even a single DNA molecule, PCR amplification has many applications in research and in the clinical laboratory for testing in genetics, oncology, infectious diseases, pharmacogenetics, and identity testing. Myriad commercial products and reagent kits are available, some with predesigned protocols that simplify the validation and implementation process for many assays, as well as manual and automated instrument platforms. Most molecular diagnostic tests incorporate target amplification by PCR, and although variations and new technologies are being introduced at a rapid pace, PCR will undoubtedly continue to play a major role in diagnostic applications.
