- W162399911 abstract "Summary and introduction This report describes the ability of human TF preparations from Mantoux positive subjects to potentiate PPD-induced inhibition of macrophage migration in mixed populations of normal guinea pig peritoneal exudate cells and human peripheral blood lymphocytes (LMMI test). The effect is dependent upon concentration of antigen (PPD) in the test system and is not regularly obtained if normal guinea pig peritoneal exudate cells are tested without being mixed with human lymphocytes. In this paper we demonstrate a method of expressing and correcting effects of HDTF upon these migration systems and an approach to determining the antigen-specificity with which potentiating effects of HDTF may occur. For some years it has been considered that cell migration inhibition systems might be suitable for detecting biological activities of human TF preparations upon lymphoid cells in vitro ( 1 ). It is now clear that HDTF contains moieties active in vitro upon lymphocytes, macrophages and polymorphs; and the analysis of effects of HDTF upon cell migration systems will be correspondingly complicated. We have chosen to study the LMMI reaction as it provides for an operationally convenient test of cellular hypersensitivity into which HDTF preparations can be introduced at various stages. In the course of this work we have therefore examined the reactivity of unsensitised guinea pig peritoneal exudate cells towards PPD and HDTF. Our results reveal both potentiating and abrogating effects of HDTF preparations and suggest that both lymphocytes and macrophages may be differently involved in these responses." @default.
