- W1625327734 abstract "Publisher Summary For the purposes of isolation of nucleoli, it is essential that a divalent ion be present in the medium for isolation of the nuclei, and hence, the sucrose-calcium procedure, for isolation of nuclei is employed. After the tissues are perfused or excised and chilled in 0.15 M NaC1 or 0.25 M sucrose, they are homogenized in 0.25 M sucrose containing 3.3 mM Ca ++ or 10 mM Mg ++ . Sucrose solutions of higher concentrations such as 2.4M are also employed. Other procedures tested for isolation of nucleoli are treatment of nuclei with citric acid and with 50% acetic acid. There are no special requirements for the mass isolation of nucleoli from the nuclei obtained by large-scale techniques as batch processes are used in this laboratory for this purpose. The sonic oscillation is carried out in a Sonifier because it can hold a somewhat greater volume than the Raytheon Sonic Oscillator. Centrifugation of the sonicate, which is layered over 0.88M sucrose is carried out in the rotor of the RC-3 centrifuge, which contains 4 swinging buckets, each of which has a volume of 650 ml." @default.
