- W162785462 abstract "Publisher Summary This chapter explores the ampliative effect, on mouse spleen cells, of isolated components of human dialyzable leukocyte lysate (DLL) 24 h after injection into KLH-sensitive mice. In a study described in the chapter, sequential Sephadex G-25 and Bio-Gel P-4 chromatography of DLL separated three distinct ampliative and two suppressive fractions. Further purification of one of the ampliative fractions by high-pressure, reverse phase, liquid chromatography (HPLC) resulted in the isolation of a molecule capable of augmenting the proliferative response to KLH by spleen cells of recipient mice. Partial elucidation of the chemical structure of this amplifier by proton-nuclear magnetic resonancespectroscopy and low-resolution mass spectrometry indicated a nucleoside structure similar to—but possibly distinct— thymidine. As the results indicate that a nucleoside structure is capable of amplifying the proliferative response to antigen, it would be interesting to study the immunomodulatory effects of other nucleosides. The demonstration of a comparable ampliative effect with either native thymidine or a known derivative of thymidine, however, would not establish the structural identity of δ component." @default.
