- W1651763399 abstract "This chapter discusses the disassociation of cell types from the cerebellum and their individual properties. The constituent functional units of a brain can be separated in a pure and viable form, thereby enabling their individual properties to be studied in isolation.The dissociation procedure is of short duration (1hr) and is carried out using media that are isotonic, well-oxygenated, and at physiological pH; lengthy centrifugation steps at high-gravitational forces are avoided, and the traumatic process of mechanical dispersion is facilitated by pretreatment of the tissue with trypsin.The tissue samples are sliced and incubated in media with or without enzymes prior to dissociation; the ability to obtain viable cells from incubated tissue slices depends, in the first instance, on the cells in the slices being viable. These cells readily grow and differentiate in culture, thus, allowing investigation into, for example, mechanisms underlying developmental events such as synaptogenesis. In an experiment described in the chapter, the competence of the isolated cells was evidenced by sustained glycolysis and coupled respiration, the maintenance of normal transmembrane gradients of Na+ and K+, and their ability to grow in culture. Another method for obtaining purified populations of cells is to use tissue culture techniques; by the choice of the conditions selected for the preferential survival of particular cell types, cultures greatly enriched in either interneurons or astrocytes were obtained in an earlier study." @default.
