It is known that 1,4-naphthoquinones are widely distributed in nature, and many derivatives have been isolated from plants, bacteria and fungi. Naturally occurring naphthoquinones have long been used in folk medicine, and one such compound, 2-hydroxy-1,4-naphthoquinone, the active ingredient of henna, is used as a cosmetic. Two major mechanisms of naphthoquinone toxicity have been identified in the chapter. First, some compounds of this type are powerful electrophiles that arylate tissue components, thereby severely disrupting cellular metabolism and causing cell death. The second mechanism of naphthoquinone toxicity involves redox cycling, with generation of “active oxygen” species. The latter are powerful oxidants that are known to be harmful in a variety of biological systems. Redox cycling is a general property of naphthoquinones, which may be initiated by either one- or two-electron reduction. Rates of naphthoquinone reduction by QR and rates of naphthohydroquinone autoxidation are analyzed in this chapter. The chapter explores mechanisms of naphthohydroquinone autoxidation. Two classes of naphthoquinone are also described in the chapter. Cytotoxicity of naphthoquinones in vitro is analyzed in the chapter." @default.
