- W167108519 abstract "ABSTRACT Modification of tumor immunogenicity by insertion of lipophilic molecules into the lipid phase of tumor cells has been investigated. A series of alkylcatechols including pentadecylcatechol and the natural plant oil urushiol have been selected for these studies since they are lipophilic and are highly potent hypersensitizing agents. The lipophilic properties of these compounds was demonstrated by the high uptake of 3 H-labeled pentadecylcatechol and the heptadecyl derivative heptadecylcatechol into erythrocyte membranes as well as normal and neoplastic cells. Furthermore model membrane studies indicated that incorporation of pentadecylcatechol or urushiol led to changes in membrane fluidity as determined by polarization of fluorescence from a fluorescence probe, 1,6-diphenyl-l,3,5 hexatriene. These membrane changes were also reflected in alterations in the antibody induced redistribution of β-2-microglobul in on pentadecylcatechol or urushiol treated lymphocytes. The influence of alkylcatechols on tumor immunogenicity has been approached firstly by demonstrating that treatment enhances the capacity of a weakly immunogenic rat hepatoma to induce immunity against transplanted tumor cells. Secondly, since these compounds are potent inducers of delayed hypersensitivity in humans and guinea pigs, experiments have been carried out to show that intralesional injection of pentadecylcatechol into intra-dermal grafts of the Line 10 hepatoma in sensitized guinea pigs suppresses growth of the local tumor and markedly restricts the development of regional lymph node metastases." @default.
