- W168020172 abstract "Abstract In short-time dust inhalation experiments with innocuous dusts (approximately 0×5 mg per rat lung), we have found only a little and transitory increase of the frequency of the alveolar cells in the lungs. This has been interpreted as retarded transport of the dust loaded cells to the bronchi as compared to the normally exchanged alveolar cells. Generally both should be intact and living when going to the bronchi. After similar inhalations of pure quartz dust (and also other cytotoxic SiO 2 modifications) there occurs a considerable increase in the mitosis rate in the alveolar region. There is a reaction on destroyed dust cells and this is accompanied by reactive changes to the intraalveolar cell debris in the lung interstices. These changes would imply erosion of the tissue barrier between the alveolar space and interstice. This reaction would make it possible for quartz particles to “penetrate” into the interstices and to go into the regional lymphatic nodes. At the same time, the bronchial dust elimination of the quartz is considerably reduced. Treatment with polyvinylpyridine- N -oxide will prevent all these changes, related to cell necrosis, which otherwise have been caused by inhalation of quartz and other dust cell destroying SiO 2 modifications. After intraperitoneal dust injections (50 mg per rat) the transport of dust particles within the interstitial tissue to the lymphatic nodes depends on several factors: the chief factor is that dust particles are set free which is possible only through necrosis of dust cells, as constantly observed in the “silicotic tissue reaction”. Further, size and form and also surface properties of the dust particles influence the rate and velocity of the transport of particles on the lymphatic passage. We have also found a different dust deposition capacity in the lymphatic nodules which is limited in relation to the grade of cytotoxicity of the dust particles. In experiments with polyvinylpyridine- N -oxide by subcutaneous injection, a different dose response relationship is found with the individual SiO 2 modifications. On the other hand, after adsorption of PNO on the SiO 2 surfaces before dust injection, there appears to be no direct relation between the coating of the surfaces and the protective effect. We may conclude therefore that the polyvinyl-pyridine- N -oxide protects not by means of a coating of the SiO 2 surfaces, but rather acts on the dust cell itself." @default.
