- W169945519 abstract "S. cerevisiae , contains two enolase structural genes (ENO1 and ENO2) per haploid genome. ENO1 gene is interesting because of heat shock response and regulation of glycolytic gene expression in yeast. The chapter describes the method of determining a positive regulatory region of ENO1 gene. A 352 bp region preceding ENO1 initiation codon is chemically synthesized according to the sequence reported, and its promoter activity is examined by constructing gene fusion with E. coli lacZ. The region, in a study described in the chapter, showed only a weak promoter activity, and the activity was greatly enhanced by joining the further upstream region ranging from –723 to –353, which was isolated from the chromosomal DNA. To examine whether the expression of the ENO1-lacZ fusion depends on the state of its existence, a single-copy (YCp) and integration type (YIp) plasmids carrying the same ENO1-lacZ fusion are constructed and the level of βGal in three different states were compared. To know whether the 86-bp alone is sufficient for the expression of ENO1, plasmids containing this region immediately upstream of the TATA box was constructed. To analyze the function of the positive regulatory region of ENO1 on the other genes, a gene fusion of PHO5, encoding repressible acid phosphatase and lacZ was constructed and substituted for the Upstream Activation Sequence of PHO5, the regulatory region of ENO1." @default.
