Summary Sirolimus is an immunosuppressive drug with a mechanism of action unlike that of tacrolimus and cyclosporine, and an effectiveness similar to the latter. It has been demonstrated to reduce the incidence of acute rejection in renal transplantation at 6 and 12 months when in combination with cyclosporine and corticoids; however, no significant change has been observed regarding both graft and patient survival. Most commonly encountered adverse events are hypertriglyceridemia, hypercholesterolemia, thrombocytopenia, and leukopenia. It is currently under study for the management of acute rejection in other transplant types, as a rescue therapy for rejection refractory to other treatments, in chronic rejection, for autoimmune diseases, and as an anti-tumour agent." @default.
