- W170622777 abstract "Saccharomyces cerevisiae accumulates {sup 64}Cu into two copper-binding pools which are distinguished by their exchange lability with respect to added extracellular copper. Cultures grown at medium (Cu{sup 2+}) {le} 1 {mu}M accumulate {sup 64}Cu only into a non-exchangeable pool. This pool was localized to the intracellular space by analysis of the distribution of {sup 64}Cu in protoplasts versus the periplasmic space and cell wall. The majority of the copper in this pool is bound to copper metallothionein and Cu,Zn superoxide disbutase. An exchangeable cellular copper-binding pool was evident in cells grown at medium (Cu{sup 2+}) {ge} 2 {mu}M. Kinetic experiments showed that {sup 64}Cu partitioned into the intracellular, non-exchangeable and exchangeable pools at a constant ratio. The exchangeable pool was localized to the cell wall and shown to be metabolically inert. Zymolyase treatment of cells released a large Mr{sup 64}Cu-binding species from the cell wall which eluted as a void volume peak on Superose 12. The metal in this fraction was exchangeable in vivo and in vitro. However, when labelled in vitro the corresponding chromatographic fraction obtained from copper naive cells bound {le} 30% of the {sup 64}Cu bound in vivo suggesting that the cell wall-associated {sup 64}Cu-complex originated withinmore » the cell. The exchangeable pool of {sup 64}Cu was absent in sulfate-free cultures and in strains mutant in the reductive assimilation of SO{sub 4}{sup 2{minus}}, and was repressed by Met and SAM, the end-products of that pathway. The data suggest that the wild type strains of S. cerevisiae respond to medium (Cu{sup 2+}) = 2-20 {mu}M by depositing a constant fraction of newly arrived copper on or in the cell wall in association with a sulfate assimilation intermediate.
