- W1712854766 abstract "Publisher Summary The content in mitochondria of various enzymes can be described in an enzyme profile. The profile summarizes the data in a graph, which is particularly designed for comparing the equipment of the mitochondria from various organs. In most cases, the enzyme profile corresponds to an enzyme activity pattern. The measure for the content is, in this case, the activity of the enzymes as found in a crude extract or sediment of the mitochondria. They include the dehydrogenases, phosphate transferases, transaminases, and so on. In principle, the enzyme activity can be converted to a molar content by the turnover number. In this case, however, the kinetic properties of the enzyme in the crude extract must agree with those of the isolated enzyme. The enzyme activity, in particular of regulatory enzymes, might be modified in the crude extract. Among the mitochondrial enzymes this concerns, for example, glutamate dehydrogenase and DPN-specific isocitrate dehydrogenase, the activity of which can be influenced by various modifiers, possibly present in the extract. The enzyme activity test can be perturbed by other enzymes in a crude extract, such as by hydrolases, isomerases, which may compete for the substrate. Careful control of these perturbations must be assured. The chapter also discusses the preparation of mitochondria, extraction, fractional extraction, and evaluation of enzyme profiles." @default.
