- W17242020 abstract "The water relations of Geotrichum candidum were characterized in an attempt to explain a previously reported relation [Baudoin & Eckert (1982), Phytopathology72, 1592–1597] between the water potential of lemon fruits and their susceptibility to decay caused by this fungus. G. candidum was grown in liquid nutrient media amended with solutes (K.C1, mannito], or polyethylene glycol 400) to achieve osmotic potentials from −0·5 to −2·5 MPa (1 MPa=10 bars). With either mannitol or polyethylene glycol 400 as an osmoticum, osmotic potential did not significantly affect growth of G. candidum, whereas KC1 at −2·5 MPa reduced growth to 57% of that at −0·5 MPa. Production of extracellular polygalacturonase (PG) was strongly influenced by osmotic potential. With all three osmotica, PG production decreased rapidly with decreasing osmotic potential, so that at −2·5 MPa, PG production was only about 1% of that at −0·5 MPa. The osmotic potential of PG assay mixtures was manipulated with sucrose, glycerol, or mannitol. The effect of osmotic potential on PG activity was minor; PG activity decreased slowly or not at all as the osmotic potential of the assay mixtures decreased from −0.5 to −2.5 MPa. The effect of osmotic potential on PG production in vitro may partially account for differences in susceptibility of lemons of different water potentials to sour rot." @default.
