• W17370385 abstract "Abstract Lipocortins are a family of proteins whose major function is to inhibit phospholipase A2, a key enzyme involved in the production of inflammatory mediators such as prostaglandins, leukotrienes and paf-acether. The mechanism of action of anti-inflammatory steroids is in part attributed to the synthesis and/or release of lipocortins. These proteins are thought to be regulated by phosphorylation involving tyrosine kinases such as the EGF receptor and the retrovirally coded protein kinase pp60 src , as well as the serine/threonine protein kinase C. Recently lipocortins were identified as members of a new family of calcium-dependent / phospholipid binding proteins including other proteins found in the literature under the names calelectrin, endonexin, calpactin, chromobindin, synhibin, calcimedin, and annexin. The comparison of available amino acid sequences showed that all these proteins contain a conserved consensus sequence of 17 amino acids. Recent kinetic studies revealed a substrate concentration-dependent inhibition of the enzyme by lipocortin involving a calcium-dependent binding to the phospholipid rather than an enzyme-inhibitor interaction. Whatever the mechanism of inhibition, these proteins are able to exert an anti-inflammatory effect in an animal model, but it is not yet known whether they can reproduce all the effects induced by anti-inflammatory steroids." @default.
