- W173832864 abstract "Our understanding of biological processes at the molecular level is, in large part, a direct consequence of knowledge obtained over the past three decades on the three-dimensional structures of biological molecules. Thus, the determination of the double-helical structure of DNA by Crick, Franklin, Watson and Wilkins led directly to the rationalization of much of genetics, and ultimately to the ability to manipulate and dissect genes at the molecular level. The knowledge that specific cellular genes, when activated by, for example, point mutations or translocations, can give rise to tumorigenic events provides for the first time the possibility of a totally specific target for chemotherapy. In order to exploit this information, it will be necessary to design drugs which interact with DNA with complete sequence-specificity; this, in turn, will require an understanding of the detailed three-dimensional structure and dynamics of these oncogene targets. This chapter describes the role of molecular modelling in the elucidation of the three-dimensional structure of biomolecules, in the analysis of the relationship between structure and function, and in the evaluation and design of drugs that can interact with DNA, proteins or enzymes." @default.
