- W1741680335 abstract "OBJECTIVE:We characterized the microglial cellular populations present in the cerebrospinal fluid (CSF) of multiple sclerosis (MS) patients and healthy controls (HC) and we determined their gene expression profile by microarray.BACKGROUND:MS is a debilitating immunologic disorder characterized by inflammation in the central nervous system (CNS). The CNS lesions of MS patients contain several immune cell subsets, including T cells, B cells, plasma cells, and macrophages; however, the role of monocytes and resident microglia (mononuclear phagocytes) in the CNS is unclear.DESIGN/METHODS:CSF and blood was obtained from healthy controls and MS patients who had been untreated for at least six months before lumbar puncture. We analyzed CSF cells from patients and controls by flow cytometry and used FACS to sort CD11c+CD14+ cells. We extracted RNA from the sorted population and analyzed its gene expression profile by microarray. To determine if monocytes in the peripheral blood share gene expression patterns with those in CSF, we enriched CD14+ cells from blood samples by magnetic cell sorting, extracted RNA, and assessed gene expression by qPCR.RESULTS:We found that the total number of monocytes and microglia analyzed as a percentage of the total number of all cells in the CSF of MS patients was significantly lower compared to the analysis of CSF derived from HC (p=0.01). Microarray analysis of CSF-derived mononuclear phagocytes showed that in MS patients, these cells express significantly lower levels of IL-10. Peripheral blood monocytes of MS patients also had lower levels of IL-10 expression as compared levels seen in HC.CONCLUSIONS:The results show that the percentage of mononuclear phagocytes is decreased in the CSF of MS patients and they have reduced expression of anti-inflammatory cytokines. This indicates that they may have a regulatory role in the CNS and that MS patients may benefit from therapies that increase IL-10 expression in mononuclear phagocytes.
