- W174240201 abstract "Mitochondrial biogenesis requires the coordinated assembly of proteins and lipids into the organelle. Mitochondria can synthesize several acidic phospholipids autonomously, including phosphatidic acid, CDP-diacylglycerol, phosphatidylglycerol, and cardiolipin. In contrast, phosphatidylcholine, phosphatidylserine, phosphatidylinositol, and sterols are synthesized in other organelles and transported to mitochondria. Imported phosphatidylserine is rapidly decarboxylated within the mitochondria to form phosphatidylethanolamine. Eukaryotic cells with defects in the mitochondrial synthesis of phosphatidylglycerol, cardiolipin, and phosphatidylethanolamine have been isolated and exhibit abnormal mitochondrial morphology or stability. The mechanisms of lipid transport to and the import into mitochondria are poorly understood. Phosphatidylcholine transport is rapid and this lipid appears to readily equilibrate between the mitochondria and endoplasmic reticulum. Phosphatidylserine transport requires special associations between the endoplasmic reticulum and mitochondria and is regulated by ubiquitination and a specific transporter for lipid movement between the outer and inner membrane. Extra-mitochondrial phosphatidylethanolamine is only poorly transported to the organelle and cannot fully replace the pool generated at the inner membrane. Phosphatidic acid, CDP-diacylglycerol, and phosphatidylinositol can also be imported into the mitochondria, but few biochemical details of these processes have been elucidated. Cholesterol transport from the outer to the inner mitochondrial membrane in mammalian adrenal cells is controlled by the sterol acute regulatory protein. Mitochondria from eukaryotic cells also export significant quantities of phosphatidylethanolamine for the biogenesis of other organelles. In yeast, as much as 70% of the total cellular phospholipid can be generated in the mitochondria. It is now clear that mitochondria play an important and dynamic role in structural phospholipid metabolism in eukaryotic cells and that they participate in both lipid import and export processes." @default.
