- W176164311 abstract "This chapter focuses on the safety of drugs used to treat major diseases in pregnant women and reviews data on the reproductive and developmental consequences of therapeutic drug exposures in human and nonhuman primates. The greatest progress in defining fetal risk from maternal drug exposure can be elucidated from studies of the teratogenic effects of xenobiotics in animal models and the scientific review of retrospective fetal outcome data in human populations. Many drugs are consumed during pregnancy to manage pre-existing chronic illnesses, to treat pregnancy-related diseases, and to cure acute bacterial and fungal infections or other conditions that occur during gestation. The major pregnancy-related medical problems that affect large numbers of women include pregnancy-induced hypertension and preeclampsia, gestational diabetes, and depression. Altered metabolism during pregnancy may lead to higher or lower exposures to potentially harmful metabolites when compared to the nonpregnant state in a species-specific manner. One of the most important methodological aspects of studies designed to investigate developmental drug exposure is the identification of the appropriate animal model. The nonhuman primate may be the most appropriate model for determining fetal exposure levels of xenobiotics and the changes in disposition of xenobiotics during pregnancy, because the physiological changes that occur during human pregnancy are poorly reflected in the rodent." @default.
