In cytochemistry, the cell can be regarded as a crude protein and nucleic acid preparation. Cells from a variety of sources can be used easily with cytochemical methods for a comparative study of a particular reaction or molecule where purity is not essential. Cytochemical methodology can be used wherever precipitable reactions are involved and locations within cells are of paramount importance. Various major discoveries in biology have been through the agency of cytochemistry. These include (1) the emphasis on the possible role of nucleic acid in direction of protein synthesis, (2) the general nature of the constancy of DNA in the haploid chromosome complement of a species following the original biochemical discovery of DNA constancy, (3) the discovery that enzymes are associated with specific structural components of cells, (4) the demonstration that DNA synthesis occurs during interphase, and (5) the demonstration of the semiconservative nature of chromosome distribution after DNA replication." @default.
