The organization of the eukaryotic cell is immensely complex with numerous compartments and microenvironments comprising distinct subsets of proteins involved in specialized functions. In the yeast, Saccharornyces cerevisiae, several membrane-bound organelles have been characterized, but within each still further layers of structural complexity can exist. Some proteins are sequestered to topologically distinct regions of the cell, such as the bud neck, apical tip, or nucleolus, whilst others function in association with multimeric protein complexes or supramolecular structures, such as the mitotic spindle. Thus to investigate fully the role of an individual protein, it is necessary both to identify its subcellular localization and to identify any biochemical interactions with other factors that may be intrinsic to its function. Antibodies raised against a specific protein can provide immensely powerful research tools for such studies. The chapter introduces the theory behind the most commonly used immunological techniques, with specific reference to their applications in yeast gene analysis. Western blotting of whole-cell extracts and the immunoprecipitation and indirect imunofluorescence of proteins is also discussed in the chapter." @default.
