- W1784452661 abstract "Publisher Summary This chapter introduces postnatally derived ventral midbrain dopamine neuron (VM) cultures as a model system for studying neurotoxicity and Parkinson's disease. The chapter also discusses different characteristics of postnatal VM cultures and examines use of these cultures for studies in dopaminergic neuronal degeneration. Techniques to maintain postnatally derived VM cultures require additional effort, due in part to the toxicity of DA itself, as well as requirements for media factors and astrocyte contact for long-term survival. The vast majority of the non-DA neurons in postnatal VM cultures are GABAergic on the basis of γ-aminobutyric acid or glutamic acid decarboxylase immunostaining. Astrocytes provide glutathione or a glutathione precursor during the conditioning, which is highly protective against L-DOPA and DA-mediated neurotoxicity in both embryonic and postnatal VM cultures. Postnatal VM cultures exhibit a high rate of DA neuron death following plating, which is apoptotic. Postnatal VM cultures can be maintained in serum free, highly defined media, and are good systems for studying potential neuroprotective factors. Postnatal VM cultures are also shown to be more restricted in their response to growth factors than embryonic cultures." @default.
