- W179667021 abstract "The effect of reperfusion of ischemic myocardium on fatty acid utilization was studied using an isolated pig heart which was subjected to 40 min of normothermic global ischemia followed by 60 min of reperfusion. (U-/sup 14/C)-palmitic acid was added to the perfusion circuit 15 min prior to ischemia. Heart biopsies and perfusates were withdrawn at regular intervals. Lipids were extracted from biopsies and products of fatty acid oxidation (/sup 14/CO/sub 2/) and incorporation of (/sup 14/C)-palmitic acid in various cellular lipid components were measured. (/sup 14/C)-palmitic acid clearance, calculated from an estimate of myocardial blood flow, dropped significantly to 21% at the onset of reperfusion as compared to 54% at the control preischemic stage. Following the ischemic insult, /sup 14/CO/sub 2/ production (a measure of ..beta..-oxidation) fell to 12% of control. With reperfusion, /sup 14/CO/sub 2/ increased and the time-activity curves leveled off after 30 min of reperfusion. (/sup 14/C)-palmitate clearance followed a similar pattern. The amounts of (/sup 14/C)-label incorporated into nonesterified fatty acids, di- and tri-glycerides, and phospholipids were 3.7%, 79.9% and 13.3% prior to ischemia; 9.7%, 76.7% and 10.8% following ischemia; and 7.4%, 80.8% and 11.5% after reperfusion.
