Under direction of SUHARSONO and UTUT WIDYASTUTI. Melastoma malabatrichum is an aluminum (Al) accumulator plant that grows well in acid soil with high level of soluble Al. Therefore, it was supposed that M. malabatrichum (Mm) has specific mechanism of Al detoxification in Al stress condition. One of the genes that play an important role in detoxification of heavy metal was metallothionein (Mt). The objective of this research was to isolate and characterize cDNA encoding metallothionein type 2 from M. malabatrichum. Total RNA was isolated from M. malabatricum’s leaves. Total cDNA was synthesized using the total RNA as template by reverse transcription. By PCR technique, the cDNA of MmMt2 was successfully isolated using Mt2 spesific primer designed from Arabidopsis thaliana (Acc.: AY037263). The MmMt2 cDNA was successfully inserted into pGEM ® -T Easy plasmid, then the recombinant plasmid was introduced into Eschericia coli DH5α. Sequencing of MmMt2 cDNA showed that MmMt2 cDNA has 246 bp encoding 81 amino acids. Enzyme restriction sites analysis showed that MmMt2 cDNA did not contain EcoR1 restriction site. Nucleotide and amino acid alignment analysis using BLAST program showed that MmMt2 cDNA was similar with complete cDNA of AtMt2A from A. thaliana (Acc.:NM_111773)." @default.
