This approach can lead to a rational or semirational approach to ligand design and can provide insights into the physical–chemical and stereostructural properties that determine peptide hormone structure—biological relationships at a 3D structural level. Many peptide hormones of biological importance are relatively small with respect to biological proteins, and as a result the degree of conformational diversity possible for these hormones in various biological environments is far greater due to the lack of a well-defined tertiary structure. Peptide hormones are produced biosynthetically from larger precursor proteins, and when formed they may adopt numerous conformations at the physiological temperature. Hence, the determination of structural elements, conformations, and dynamic properties is of critical importance to biological activity. The specific functional groups in the primary sequence that are related to specific biological effects should be considered. The relationships between the conformational properties and dynamic behavior and the measured biological activity should be comprehended. Assays can provide a sensitive probe to the structural, conformational, and dynamic requirements for a biological activity for a peptide than many of the more sophisticated biophysical experiments such as nuclear magnetic resonance spectroscopy and X-ray crystallography." @default.
