- W1830758410 abstract "Publisher Summary This chapter focuses on avocado and apple mitochondria. The preparation requires that the tissue be disrupted in isotonic sucrose under conditions, which gives reasonable yields and yet not disrupt the mitochondria. Acidity, tannin formation, and oxidation products lead to inactivation of mitochondria and release of fatty acids can cause uncoupling of oxidative phosphorylation. These difficulties are usually overcome by adding alkaline buffers, polyvinylpyrollidone, cysteine (or ascorbate), and bovine serum albumin to the isolating medium. Only soluble polyvinylpyrollidone has been used, but the use of an insoluble cross-linked form may be more advantageous. The activities of mitochondria isolated from these fruits tend to display seasonal variations, which are particularly associated with the climacteric pattern of respiration. These variations are possibly not inherent but a reflection of the ease or difficulty in isolating the desired mitochondria. Apart from problems associated with the presence of inhibiting-substances in the tissue there is sometimes a need for strong shearing forces to disrupt the tissue." @default.
