- W1838863859 abstract "This chapter introduces standard reversed-phase high performance liquid chromatography and UV-photodiode array detection (RPLC-UV) methodologies for the analysis of nucleosides and nucleoside composition in RNAs. The chromatographic protocols and standard nucleoside columns are presented and the essential requirements for the HPLC instrumentation are described. Three optimized RPLC systems are developed, with particular emphasis on—resolution, speed, or sensitivity. Three unfractionated transfer RNA (tRNA) are selected as sources of the reference nucleosides and for the assessment of the performance of chromatography. From these, a large array of nucleosides are characterized from the hydrolysates of these three reference tRNAs that are used in standardization and calibration of the method. The chapter discusses the use of adiode-array detector for the detection of nucleosides for enhancement of the reliability of nucleoside identification and accuracy of measurement, and an extended enzymatic hydrolysis protocol for the liberation of exotically modified nucleosides in transfer RNA (tRNA). A chromatographic performance test is included to reproduce nucleoside chromatography protocols on high-performance liquid chromatography (HPLC) instrumentation. This test provides information on the true gradient delay time, slope of the gradient tramps, pump pulsation, and mixing of the buffers." @default.
