Our study was aimed at assessingcurcumin labelling of characteristic pathologicaldeposits of hyperphosphorylated tau protein in brain sections from representativecases of three major classes of tauopathies:Alzheimer’s disease (AD; class 1), progressivesupranuclear palsy (PSP; class 2) and Pick’sdisease (PiD; class 3).Methods: Th e structures of interest were fi rstidentifi ed in HE-stained sections and photographed.Subsequently, the visualization ofthese structures with curcumin and AT8 immunofluorescence was assessed by sequentiallabelling and computer-assisted photographyof the same loci. Results were expressedas percentage of structures labelled.Results: Curcumin detected fi brillar tau inAD (95 %) and PSP (90 %), but not in PiD.When comparing curcumin labelling to AT8 immunofl uorescence in AD and PSP, curcuminlabelled fi brillar AT8-positive, but notnon-fi brillar AT8-immunofl uorescent structures.Curcumin also labelled extracellularfi brillar tangles remaining aft er neuronaldeath in AD and PSP, which were not visualizedby AT8 immunofl uorescence.Conclusions: Curcumin fl uorescence wasshown to diff erentiate between pathologicaltau deposits in two ways. Firstly, it preferentiallydetected tau deposits with fi brillarmorphology. Secondly, curcumin alsodiff erentiated between representative casesof main tauopathy classes: it did not revealfi brillar Pick bodies, while clearly labellingneurofi brillar tangles found in AD and PSP." @default.
